RESUMO

The present review will present the recent published results and discuss the main effects of nutrients, mainly fatty acids, on the expression of genes involved in lipid metabolism. In this sense, the review focuses in two phases: prenatal life and finishing phase, showing how nutrients can modulate gene expression affecting marbling and fatty acid profile in meat from ruminants. Adiposity in ruminants starts to be affected by nutrients during prenatal life when maternal nutrition affects the differentiation and proliferation of adipose cells enhancing the marbling potential. Therefore, several fetal programming studies were carried out in the last two decades in order to better understand how nutrients affect long-term expression of genes involved in adipogenesis and lipogenesis. In addition, during the finishing phase, marbling becomes largely dependent on starch digestion and glucose metabolism, being important to create alternatives to increase these metabolic processes, and modulates gene expression. Different lipid sources and their fatty acids may also influence the expression of genes responsible to encode enzymes involved in fat tissue deposition, influencing meat quality. In conclusion, the knowledge shows that gene expression is a metabolic factor affecting marbling and fatty acid profile in ruminant meat and diets and their nutrients have direct effect on how these genes are expressed.

Steer progeny suckled by cows fed a dried distillers grains and solubles (DDGS) diet the first 3 mo of lactation were heavier during feedlot finishing and had significantly lower marbling and larger longissimus muscles than steers suckled by cows fed a control diet (CON). These differences were profound in that progeny were managed and fed identically from weaning until finishing, and findings suggest that the suckling period established the developmental program of muscle composition. Here transcriptomes of longissimus muscle were measured by next-generation sequencing to investigate whether there were any developmental clues to the differences in marbling scores and muscle content between steers suckled by DDGS ( n = 5) vs. control (CON; n = 5) diet-fed cows during lactation. There were 809 genes differentially expressed ( P-adj<0.1) between CON and DDGS muscle. Of these 636 were upregulated and 173 downregulated in DDGS relative to CON. Overall the DDGS vs. CON muscle transcriptomic signature was promyogenic and antiadipogenic. In particular, myokines/satellite cell maintenance factors were found among upregulated (LIF, CNTF, FGFB1, EPHB1) genes. The antiadipogenic signature was typified by the upregulation of anti-inflammatory cytokines and receptors (IL1RAP, IL1RL2, IL13RA2, IL1F10), and downregulation of expression of inflammation/inflammatory cytokines and receptor (TNF, IL6R, CXCL9), which suggests a selection of differentiation pathways away from adipogenic line. The upregulation of TGFB, SPP1, and INHBA supports selection of fibroblast lineage of cells. Thus, the lactation phase of production can effect meat quality by affecting transcriptional signatures that favor myogenesis and depress inflammation.

Feeding dried distillers grains with solubles (DDGS), a feed high in fat and protein, to lactating beef cows can alter milk production and composition, resulting in improved pre-weaning growth of progeny. This alteration in milk profile may consequently alter the growth and carcass composition of the offspring after weaning. Therefore, Angus×Simmental steers (n=48) whose dams were fed one of two diets supplemented with either DDGS or soybean meal (CON) from calving to mid-lactation were placed in a feedlot to determine the effects of maternal nutrition during lactation on progeny development and carcass composition. Cow-calf pairs were allotted to two treatments at birth based on cow and calf BW, breed and age. Maternal diets were isocaloric (3.97 MJ/kg NEg) and consisted of rye hay supplemented with DDGS at 1% of BW (19.4% CP; 8.76% fat) or rye hay and corn silage supplemented with CON (11.7% CP; 2.06% fat). After conclusion of the treatments at 129 days postpartum, cow-calf pairs were comingled and managed as one group until weaning at 219 days postpartum. Steers were then transitioned to a common diet composed of 60% DDGS, 34% corn silage and 6% vitamin/mineral supplement and were placed indoors in individual pens with slatted floors. An intravenous glucose tolerance test (IVGTT) was performed 134 days after feedlot entry on 16 steers (CON, n=7; DDGS, n=9) to determine the effect of maternal diet on glucose and insulin sensitivity. Steers were slaughtered at a target BW of 645 kg. Categorical and continuous data were analyzed using the GLIMMIX and MIXED procedures of SAS, respectively. Steers from DDGS dams tended to be heavier on day 85 of feedlot finishing (P=0.09) compared with steers from CON dams. However, there were no differences in final weight, average daily gain, dry matter intake or efficiency (gain:feed, P⩾0.18). Maternal treatments did not affect progeny days on feed (P=0.15), despite a mean difference of 9 days in favor of DDGS. Glucose and insulin concentrations and area under the curve of progeny as measured by IVGTT were not affected by maternal diet (P⩾0.16). Maternal DDGS supplementation decreased marbling score (P=0.04), but did not influence carcass grading percentage or any other carcass characteristic (P⩾0.17).

Three experiments were conducted to determine the effect of increasing concentrations of a smectite clay on toxin binding capacity, ruminal fermentation, diet digestibility, and growth of feedlot cattle. In Exp. 1, 72 Angus × Simmental steers were blocked by BW (395 ± 9.9 kg) and randomly allotted to 3 treatments (4 pens/treatment and 6 steers/pen) to determine the effects of increasing amounts of clay (0, 1, or 2%) on performance. The clay was top-dressed on an 80% concentrate diet at a rate of 0, 113, or 226 g/steer daily to achieve the 0, 1, and 2% treatments, respectively. Steers were slaughtered at a target BW of 606 kg. In Exp. 2, 6 steers (596 ± 22.2 kg initial BW) were randomly allotted to the same 3 treatments in a replicated 3 × 3 Latin square design (21-d periods) to determine the effects of increasing amounts of clay on ruminal pH, VFA, and nutrient digestibility. In Exp. 3, 150 mg of clay was incubated in 10 mL of rumen fluid with 3 incremental concentrations (6 replicates per concentration) of aflatoxin B (AFB) or ergotamine tartate (ET) to determine binding capacity. During the first 33-d period, there was a quadratic effect of clay on ADG ( < 0.01) and G:F ( < 0.01), increasing from 0 to 1% clay and then decreasing from 1 to 2% clay. However, during the second 30-d period, clay linearly decreased ADG and G:F ( ≤ 0.03) and overall ADG, DMI, and G:F were not impacted ( ≥ 0.46). Clay linearly decreased marbling score ( = 0.05). Hepatic enzyme activity did not differ among treatments on d 0 or at slaughter ( ≥ 0.15). Clay linearly decreased ruminal lactate and propionate, linearly increased formate and the acetate:propionate ratio ( ≤ 0.04), and tended ( = 0.07) to linearly increase butyrate. Clay tended to linearly increase ( = 0.06) OM and CP apparent digestibility. Ruminal pH, urine pH, and other digestibility measures did not differ among treatments ( ≥ 0.15). Clay was able to effectively bind AFB and ET at concentrations above the normal physiological range (52 and 520 µg/mL), but proportional adsorption was decreased to 35.5 and 91.1% at 5,200 µg/mL ( < 0.01) for AFB and ET, respectively. In conclusion, clay effectively binds ruminal toxins, decreases ruminal lactate, and improves performance only during adaptation to a high-concentrate feedlot diet.

Angus × Simmental cows ( = 48, BW = 594 kg, BCS = 5.26, age = 2.7) pregnant with male fetuses were used to determine the effect of selenium source during the last 80 d of gestation and first 108 d of lactation on progeny feedlot performance. At 203 d of gestation, cows were blocked by BW, breed composition, and calf sire and randomly allotted to 1 of 3 treatments: no supplemental Se, 3 mg/d inorganic Se (sodium selenite), and 3 mg/d organic Se (Sel-plex). Maternal diets were formulated to contain 10.4% CP and 0.90 Mcal/kg NE during gestation and 12.1% CP and 1.01 Mcal/kg NE during lactation. Basal diets contained 0.07 and 0.11 mg/kg Se for gestation and lactation diets, respectively. Diets were fed daily as a total mixed ration, and no additional Se, 3 mg/d Se as sodium selenite, or 3 mg/d Se as Sel-Plex were top-dressed daily. Treatment diets were fed through 108 d postpartum (DPP). At 108 DPP cow-calf pairs were commingled until weaning at 210 DPP. At 28 d postweaning, steers ( = 47, BW = 301 kg) were placed in individual pens and fed a diet formulated to provide 13.9% CP and 1.24 Mcal/kg NE. No supplemental Se was fed; however, basal Se concentration was 0.10 mg/kg. The diet was delivered as a total mixed ration once daily. Steers were slaughtered at a target BW of 625 kg. Steers from cows supplemented with organic Se tended to enter the feedlot heavier ( = 0.06) than steers from cows supplemented with inorganic Se. There was no difference in ADG among treatments ( ≥ 0.73), but steers from organic Se cows tended to spend fewer days on feed compared to steers from inorganic Se cows ( = 0.09). Steers from organic Se cows had a greater overall DMI compared to steers from inorganic Se cows ( = 0.04), but there was no difference in overall G:F ( = 0.49). Dressing percentage was greater for steers from cows fed no Se compared with steers from cows fed either inorganic or organic Se ( = 0.03). Maternal Se source had no effect on HCW, back fat, percentage KPH, LM area, yield grade, marbling score, or quality grade distribution ( ≥ 0.17) of progeny. In conclusion, maternal supplementation with organic Se appears to have a long-term benefit on intake of steer progeny and may result in improvements in growth that could decrease days on feed.

Angus × Simmental cows ( = 48; BW = 595 ± 17.4 kg, BCS = 5.26 ± 0.05, and age = 2.3 ± 0.07 yr), pregnant with male fetuses, were used to determine the effect of Se source during the last 80 d of gestation and first 108 d of lactation on cow and calf performance. At 203 d in gestation, cows were blocked by BW, breed composition, age, and calf sire and randomly allotted to organic Se, inorganic Se, or no Se treatments. Diets contained corn silage, corn stover, haylage, dried distillers' grains with solubles, and minerals and were formulated to contain 10.4% CP and 0.90 Mcal/kg NEg during gestation and 12.1% CP and 1.01 Mcal/kg NEg during lactation. Diets were fed daily as a total mixed ration and none, 3 mg/d Se as sodium selenite, or 3 mg/d Se as Sel-Plex were top-dressed daily. At 68 d postpartum (DPP), milk production was calculated using the weigh-suckle-weigh procedure and a milk sample was collected to determine composition. At 108 DPP, cow-calf pairs were commingled until weaning at 210 DPP. Cow BW and BCS ( ≥ 0.56) did not differ between treatments at any time point during the study. Milk production, milk fat, and total solids ( ≥ 0.38) did not differ among treatments. Milk protein tended to increase in cows fed inorganic Se compared with cows fed organic Se ( = 0.07) and milk lactose tended to be greatest in cows fed organic Se ( = 0.10). Conception to AI and overall pregnancy rates did not differ between treatments ( ≥ 0.39). Calf weights and ADG did not differ through 108 DPP ( ≥ 0.77) or for the preweaning period ( ≥ 0.33). Plasma Se concentration was adequate for all cows and did not differ among treatments for cows ( ≥ 0.37) or calves ( ≥ 0.90). Liver Se concentrations in cows fed inorganic or organic Se were greater than in control cows ( < 0.01). Longissimus muscles biopsies taken from progeny at 108 DPP also did not differ between treatments ( = 0.45). In conclusion, dietary Se source did not affect cow performance, milk production, or reproductive efficiency. Organic Se decreased milk protein and increased milk lactose but did not alter preweaning performance of progeny from Se-adequate cows.

The objectives of this study were to test the interaction of breed (Nellore or Angus) and diet (whole shelled corn [WSC] or ground corn [GC] with silage) on growth performance, carcass characteristics, and ruminal pH of young bulls. Thirty-six bulls (18 Nellore and 18 Angus) with the range in age of 18 to 22 mo and BW of 381 ± 12 kg were used in a completely randomized design experiment with a 2 × 2 factorial arrangement of treatments (2 breeds and 2 diets). Experimental diets (DM basis) included 1) a GC diet containing 30% corn silage and 70% GC- and soybean meal-based concentrate and 2) a WSC diet containing 85% WSC and 15% of a soybean meal- and mineral-based pelleted supplement. An additional 8 bulls were slaughtered at the beginning of the experimental period for determination of initial carcass weight. The treatments were Nellore fed the GC diet, Nellore fed the WSC diet, Angus fed the GC diet, and Angus fed the WSC diet. Greater DMI ( < 0.01), ADG ( < 0.01), and G:F ( < 0.01) were observed in Angus bulls compared with Nellore bulls, regardless of diet. Lower average ruminal pH ( = 0.04), maximum ruminal pH (P = 0.02), and DMI ( < 0.01) were observed in bulls fed the WSC diet than in those fed the GC diet. In addition, bulls fed the WSC diet had greater G:F ( < 0.01). The WSC diet led to greater variation in DMI compared with the GC diet ( < 0.01). Omasum and large intestine percentage was affected by diets only in the Angus breed ( < 0.02) and were greater when bulls were fed the GC diet. The WSC diet without forage may be useful for feedlots because this diet promoted greater G:F than the GC diet, regardless of breed. However, special care must be exercised in feed management during adaptation and throughout the feeding of Nellore animals to avoid digestive disorders and fluctuations in DMI.

Multiparous Angus×Simmental cows (n=54, 5.22±2.51 years) with male progeny were fed one of two diets supplemented with either dried distillers grains with solubles (DDGS) or soybean meal (CON), from calving until day 129 postpartum (PP) to determine effects of excess protein and fat on cow performance, milk composition and calf growth. Diets were formulated to be isocaloric and consisted of rye hay and DDGS (19.4% CP; 8.76% fat), or corn silage, rye hay and soybean meal (11.7% CP; 2.06% fat). Cow-calf pairs were allotted by cow and calf age, BW and breed. Cow BW and body condition score (BCS; P⩾0.13) were similar throughout the experiment. A weigh-suckle-weigh was performed on day 64 and day 110±10 PP to determine milk production. Milk was collected on day 68 and day 116±10 PP for analysis of milk components. Milk production was unaffected (P⩾0.75) by dietary treatments. Milk urea nitrogen was increased at both time points in DDGS compared with CON cows (P<0.01). Protein was decreased (P=0.01) and fat was increased (P=0.01) in milk from DDGS compared with CON cows on day 68 PP. Compared to CON, DDGS decreased medium chain FA (P<0.01) and increased long chain FA (P<0.01) at both time points. Saturated FA content of milk was decreased (P<0.01) at both time-points in DDGS compared with CON cows, which resulted in an increase (P<0.01) in monounsaturated and polyunsaturated FA, including cis-9, trans-11 conjugated linoleic acid. Daily gain of the DDGS calves was increased (P=0.01) compared with CON calves, resulting in heavier BW on day 129 (P=0.01). Heavier BW of DDGS calves was maintained through weaning (P=0.01). Timed-artificial insemination (TAI) rates were greater for cows fed DDGS compared with cows fed CON (P<0.02), but dietary treatment had no effect on overall pregnancy rates (P=0.64). In summary, feeding DDGS to lactating beef cows did not change cow BW or BCS, but did improve TAI rates and altered milk composition compared with CON. As a result, male progeny from cows fed DDGS during lactation had greater average daily gain and were heavier at day 129 and at weaning compared with male progeny from cows fed a control diet.

This study compared postweaning growth, puberty attainment, and reproductive processes of female progeny (n = 33) born to Angus-Simmental beef heifers treated with either a control diet or a diet in which dried distiller's grains with solubles (DDGS) were fed as an energy source during late gestation and early lactation. From 192 d of gestation through 118 ± 4 d in lactation, dams were fed either a corn silage­based control diet (CON) orcorn residue with DDGS, where DDGS were supplemented as an energy source (DG). Diets were formulated to provide similar daily NEg between diets, but CP requirements were drastically exceeded in the DG treatment. Heifer progeny (n = 33) were weaned, commingled at 191 ± 4 d of age, and similarly managed for the remainder of the project. Heifer BW and blood samples for progesterone assessment to determine onset of puberty were collected weekly beginning at weaning. At 255 ± 4 d of age, a single follicular wave was mapped via ultrasonography in 10 prepubertal heifers per treatment. Prepubertal antral follicle count and ovarian size were determined at 253 ± 4 d of age. Hip height was recorded at 213,297, and 437 ± 4 d of age. Estrous synchronization and AI was initiated at 447 ± 4 d of age. Binary data were analyzed with the GLIMMIX procedures of SAS and all other data were analyzed with the MIXED procedures of SAS. Progeny from DG-treated dams tended to be heavier (P = 0.08) than progeny from CON-treated dams from weaning until breeding. In addition, DG progeny had a greater (P < 0.01) frame score than CON throughout the developmental period. Ovarian size, antral follicle count, and follicular growth parameters did not differ between treatments. Age at puberty did not differ between CON (303 ± 10 d) and DG (320 ± 10 d) progeny; however, BW at puberty was greater (P = 0.01) for DG (326 ± 7 kg) than CON (298 ± 8 kg) progeny. Pregnancy rates to AI were greater (P = 0.05) in DG progeny (70.6%) than CON (33.3%), but overall breeding season pregnancy rate did not differ (P = 0.97). Moreover, rate of dystocia in female progeny at first parturition and grand-offspring birth BW did not differ due to treatment (P ≥ 0.74). In summary, feeding DDGS as an energy source during late gestation and early lactation to first-parity heifers resulted in female progeny with greater skeletal growth that were heavier at onset of puberty and had increased AI pregnancy rates.

Degree of unsaturation of fatty acids, which is influenced by lipid source and level of metabolism in the rumen, is a major determinant in how dietary lipids affect genes that regulate beef marbling. A total of 28 Red Norte bulls with an initial live weight of 361±32 kg (P>0.05) were used in a completely randomized experimental design to analyze the expression of genes that are involved in lipid metabolism in the longissimus dorsi (LD) when diets contained soybean grain or rumen-protected fat, with or without monensin. Treatments were arranged as a 2×2 factorial, with 4 treatments and 7 replicates per treatment. Half of the animals that received soybean or rumen-protected fat were supplemented with 230 mg head(-1) d(-1) of monensin. Gene expression was analyzed by reverse-transcription quantitative PCR (RT-qPCR). Expression of sterol regulatory element-binding protein-1c (SREBP-1c) in the LD muscle was not affected by lipid source or monensin (P>0.05). There was an interaction effect (P<0.05) between lipid source and monensin for peroxisome proliferator-activated receptor α (PPAR-α) and stearoyl-CoA desaturase (SCD) expression, where greater gene expression was found in animals fed soybean plus monensin and the lower gene expression was found in animals fed rumen-protected fat plus monensin. Expression of lipoprotein lipase (LPL) and fatty acid-binding protein 4 (FABP4) were greater (P<0.05) in the LD muscle of animals fed soybean. Monensin had no effect on LPL and FABP4 expression when soybean without monensin was fed, but when rumen-protected fat was fed, monensin increased LPL expression and decreased FABP4 expression (P<0.05). Linoleic and arachidonic acids had negative correlations (P<0.05) with the expression of PPAR-α, SCD, FABP4, and LPL genes. PPAR-α gene expression was not correlated with SREBP-1c but was positively correlated with SCD, FABP4, LPL, and glutathione peroxidase (GPX1) gene expression (P<0.001). Lipid sources and monensin interact and alter the expression of PPAR-α, SCD, acetyl CoA carboxylase α (ACACA), LPL, FABP4, and GPX1. These changes in gene expression were most associated with arachidonic and α-linolenic acids and the ability of lipid sources and monensin to increase these fatty acids in tissues.

The objectives of this study were to determine the effects of feeding dried corn distillers grains (DDGS) or modified wet corn distillers grains (MDGS) with or without CaO treatment to feedlot steers on 1) growth performance and carcass characteristics and 2) diet digestibility, pattern of intake, and meal distribution. In Exp. 1, steers (n = 139; average initial BW = 336 ± 75 kg) were used in a randomized complete block design. Treatments were arranged in a 2 × 2 factorial design, and pens were randomly allotted to 1 of the 4 dietary treatments (DM basis): 1) 50% DDGS untreated, 2) 48.8% DDGS treated with 1.2% CaO, 3) 50% MDGS untreated, or 4) 48.8% MDGS treated with 1.2% CaO. The remainder of the diet was corn husklage, dry rolled corn, and vitamin and mineral supplement. In Exp. 2, fistulated steers (n = 8; average initial BW = 540 ± 250 kg) were used in a replicated 4 × 4 Latin square design with the same dietary treatments as in Exp. 1. There was no interaction (P ≥ 0.14) between distillers grains plus solubles (DGS) and CaO inclusion for DMI, ADG, final BW, or USDA yield and quality grades. However, steers fed CaO-treated DGS had decreased (P < 0.01) DMI, regardless of DGS type. Because CaO treatment decreased DMI without affecting (P = 0.66) ADG, steers fed CaO-treated DGS had increased (P < 0.01) G:F compared to steers not fed CaO. The variation in DMI found in this experiment could be explained by differences in meal size and distribution. Steers fed CaO-treated DGS ate a similar (P = 0.36) number of meals but ate smaller (P < 0.01) meals. No effects (P ≥ 0.55) of CaO treatment or its interaction with DGS type were found for apparent total tract DM or NDF digestibility. However, steers fed MDGS had increased (P < 0.01) NDF digestibility compared to steers fed DDGS. In conclusion, CaO treatment of DGS improved feed efficiency when DGS-based diets were fed but did not improve digestibility.

To evaluate the effects of ß-carotene (ßC) on performance and carcass characteristics, 112 crossbred calves were allotted to 4 treatments consisting of retinyl palmitate (RP) supplemented at 2200IU/kg, and synthetic ß-carotene supplemented at 1 (SßC1X), 5 (SßC5X), or 10 (SßC10X) times RP. Compared to RP, SßC1X did not impact performance (P≥0.25) or plasma concentrations of retinoids or carotenoids (P≥0.26). Increasing dietary ßC increased (P≤0.04) plasma concentrations of RP and all of the isomers of ßC, but did not affect weight, gain, or gain:feed (P≥0.24). Marbling and fat thickness did not differ between RP and SßC1X or because of amount of ßC (P≥0.35). Lightness (L*) of LM decreased with increasing concentration of SßC (linear; P=0.01). Yellowness (b*) increased (P=0.04) and redness (a*) tended to increase (P=0.09) in subcutaneous fat as SßC amount increased (quadratic).

Two experiments were conducted to determine the effect of increasing dietary CaO on ruminal fermentation, diet digestibility, performance, and carcass characteristics of feedlot steers fed 60% dried distillers grains with solubles ( DDGS: ). In Exp. 1, 120 steers were allotted by weight (355 ± 7.9 kg) to 1 of 4 treatments containing 60% DDGS, 20% corn silage, 13.5 to 14.4% ground corn, 4% supplement, and 0 to 2.5% limestone on DM basis to determine the effects of CaO on performance and carcass characteristics. Treatments consisted of 0, 0.8, 1.6, or 2.4% CaO inclusion in the diet (DM basis), with CaO replacing limestone. Steers were slaughtered at a target BW of approximately 641 kg. In Exp. 2, 4 steers (initial BW = 288 ± 3 kg) were randomly allotted to the same diets in a 4 × 4 Latin square design (14-d periods) to determine the effects of CaO on ruminal pH, VFA, and nutrient digestibility. Statistical analyses were conducted using the MIXED procedure of SAS. Inclusion of CaO at 0.8, 1.6, and 2.4% increased ADG by 5.0, 3.9, and 0%, respectively, compared to 0% CaO (quadratic; P = 0.03). Intake was linearly decreased (P = 0.04) and G:F was linearly increased (P = 0.02) by CaO inclusion. Dressing percentage increased as CaO increased from 0 to 1.6% and then decreased for 2.4% CaO (quadratic; P < 0.01). In Exp. 2, steers fed 0% CaO had the greatest prefeeding ruminal pH, steers fed 0 and 0.8% CaO exhibited the most rapid postfeeding decline in ruminal pH, and steers fed 2.4% CaO exhibited a relatively stable ruminal pH throughout the 24-h period (treatment × time; P ≤ 0.01). Acetate, butyrate, and total VFA concentrations increased linearly (P ≤ 0.05) at 0, 3, 6, and 12 h postfeeding with increasing CaO. Propionate at 3 h postfeeding increased from 0 to 1.6% CaO and decreased from 1.6 to 2.4% CaO (quadratic; P = 0.10). Urine pH increased linearly (P ≤ 0.01) while urine output and urine ammonia decreased linearly (P ≤ 0.05) as CaO inclusion increased. Apparent NDF digestibility tended to increase (P = 0.07) and ADF digestibility did (P = 0.01) increase linearly with increasing concentrations of CaO. In conclusion, CaO improved ruminal pH variation, increased fiber digestibility, and decreased metabolic acid load in cattle fed 60% DDGS-based diets. Inclusion of CaO up to 1.6% was effective in improving performance of feedlot cattle.

The manipulation of acid-base balance has been extensively investigated as a means of manipulating Ca homeostasis and managing milk fever in dairy cows. A low dietary cation anion difference (DCAD) increases urinary Ca, blood-ionized Ca, and responsiveness to Ca-homeostatic hormones. Very little attention has been focused on the possibility of using a low dietary DCAD to increase muscle Ca availability, calpain activity, and meat tenderness of beef cattle. Thus, 90 Angus × Simmental crossbred steers were allotted by weight (590.1 ± 2.4 kg) and breed composition (% Simmental) to 3 treatments (6 pens/treatment, 5 steers/pen) to evaluate the effects of DCAD on beef tenderness. Treatments were initiated 2 wk before slaughter and consisted of 3 DCAD (mEq/100 g) treatments: -16, 0, and +16. Basal diets (DM basis) were 62 to 64% corn, 6 to 9% soybean meal, and 20% corn silage, and were formulated to contain similar concentrations of protein, energy (NEm; NEg), and minerals, with the exception of sodium and chlorine. A commercial chloride ion supplement (PASTURChlor, West Central, Ralston, IA) was added to diets to decrease DCAD and sodium bicarbonate was added to diets to increase DCAD. Performance before initiation of the study did not differ among treatments (P > 0.22). Urine pH did not differ at the initiation of the study (P > 0.57), but did increase at a decreasing rate on d 7 (6.37, 7.69, 8.13) and d 14 (5.68, 7.66, 8.03) of the study as DCAD increased from -16 to 0 to +16, respectively (quadratic, P < 0.02). Gain and gain:feed responded quadratically to DCAD (P < 0.01), increasing from -16 to 0 DCAD and decreasing from 0 to +16 DCAD. Hot carcass weight, dressing percent, fat thickness, LM area, yield grade, marbling score, quality grade distribution, 48 h muscle pH, and Ca content of muscle did not differ among treatments (P > 0.16). In addition, DCAD did not affect Warner-Bratzler shear force among treatments after 7 and 21 d of aging (P > 0.23). Although urine pH was decreased by feeding a -16 DCAD diet, Ca influx into the LM and beef tenderness were not affected by altering the DCAD in finishing beef cattle diets.

The main goal of this study was to develop tools for genetic selection of animals producing milk with a lower concentration of saturated fatty acids (SFA) and a higher concentration of unsaturated fatty acids (UFA). The reasons for changing milk fatty acid (FA) composition were to improve milk technological properties, such as for production of more spreadable butter, and milk nutritional value with respect to the potentially adverse effects of SFA on human health. We hypothesized that genetic polymorphisms in solute carrier family 27, isoform A6 (SLC27A6) fatty acid transport protein gene and fatty acid binding protein (FABP)-3 and FABP-4 (FABP3 and FABP4) would affect the selectivity of FA uptake into, and FA redistribution inside, mammary epithelial cells, resulting in altered FA composition of bovine milk. The objectives of our study were to discover genetic polymorphisms in SLC27A6, FABP3, and FABP4, and to test those polymorphisms for associations with milk FA composition. The results showed that after pairwise comparisons between SLC27A6 haplotypes for significantly associated traits, haplotype H3 was significantly associated with 1.37 weight percentage (wt%) lower SFA concentration, 0.091 lower SFA:UFA ratio, and 0.17 wt% lower lauric acid (12:0) concentration, but 1.37 wt% higher UFA and 1.24 wt% higher monounsaturated fatty acid (MUFA) concentrations compared with haplotype H1 during the first 3 mo of lactation. Pairwise comparisons between FABP4 haplotypes for significantly associated traits showed that haplotype H3 was significantly associated with 1.04 wt% lower SFA concentration, 0.079 lower SFA:UFA ratio, 0.15 wt% lower lauric acid (12:0), and 0.27 wt% lower myristic acid (14:0) concentrations, but 1.04 wt% higher UFA and 0.91 wt% higher MUFA concentrations compared with haplotype H1 during the first 3 mo of lactation. Percentages of genetic variance explained by H3 versus H1 haplotype substitutions for SLC27A6 and FABP4 ranged from 2.50 to 4.86% and from 4.91 to 7.22%, respectively. Tag single nucleotide polymorphisms were identified to distinguish haplotypes H3 of SLC27A6 and FABP4 from others encompassing each gene. We found no significant associations between FABP3 haplotypes and milk FA composition. In conclusion, polymorphisms in FABP4 and SLC27A6 can be used to select for cattle producing milk with lower concentrations of SFA and higher concentrations of UFA.

Angus × Simmental steers (n = 210; initial BW 314 ± 11 kg) were separated into heavy and light BW blocks and allotted evenly by BW to 6 treatments (3 heavy and 2 light pens per treatment) to determine the effect of supplemental vitamin D3: 0 IU (no D), 250,000 IU for 165 d (long-term D), or 5 × 10(6) IU for 10 d (short-term D) on performance, carcass traits, vitamin D metabolites, and meat tenderness in steers fed either 0 (NZ) or 8.38 mg/kg zilpaterol hydrochloride (ZH) daily for 21 d. Placebo or ZH was added to the diet 24 d, and short-term D was added 13 d before slaughter. Vitamin D3, ZH, and placebo were all removed from the diet 3 d before slaughter. Steers fed ZH tended to have improved overall G:F compared with steers not fed ZH (P < 0.09). Overall performance was not affected by long-term D, with or without ZH (P = 0.11) compared with no D, with or without ZH. Short-term D decreased final BW, ADG, and G:F (P = 0.04) compared with no D, when ZH was not fed. Zilpaterol hydrochloride increased HCW, dressing percentage, and LM area (P < 0.01); and decreased fat thickness, yield grade, and marbling (P < 0.03). Carcass traits were not impacted by long-term D without ZH (P > 0.13), but long-term D with ZH decreased percentage KPH (P < 0.02). Compared with no D, short-term D tended to decrease HCW (P < 0.07), decreased fat thickness (P < 0.01), and tended to increase dressing percentage (P < 0.10) when ZH was not fed, yet did not impact carcass traits when ZH was fed (P < 0.13). Feeding ZH tended to decrease (P < 0.09) LM 1,25-dihydroxyvitamin D3 [1,25(OH)2D3]. The long-term D treatment increased LM vitamin D3 and 25-hydroxyvitamin D3 (25OHD3) 18- and 5-fold, respectively, when ZH was not fed (P < 0.04) and increased LM 25OHD3 by 4-fold when ZH was fed (P < 0.01). Short-term D increased LM vitamin D3 and 25OHD3 by 52- and 9-fold, respectively, when ZH was not fed (P < 0.01), and by 24- and 9-fold, respectively, when ZH was fed (P < 0.01). Also, short-term D increased LM 1,25(OH)2D3 by 2-fold (P < 0.04) when ZH was fed. Warner-Bratzler shear force (WBSF) was greater for ZH steaks than non-ZH steaks at 7, 14, and 21 d postmortem aging (P < 0.01). Vitamin D did not reduce WBSF (P = 0.18). When ZH was fed, long-term D tended to increase WBSF in steaks aged 21 d (P = 0.06). In conclusion, ZH improved carcass leanness and decreased tenderness, and vitamin D feeding increased vitamin D3 metabolites in LM, but did not improve tenderness in steers fed ZH.

Two hundred and ten Angus × Simmental steers (initial BW 314 ± 11 kg) were separated into heavy and light BW blocks and allotted evenly by BW to 6 treatments (3 heavy and 2 light pens per treatment) to determine the effect of supplemental vitamin D3: 0 IU (no D), 250,000 IU for 165 d (long-term D), or 5 × 10(6) IU for 10 d (short-term D) on plasma and muscle calcium concentrations and gene expression in steers fed either 0 (NZ) or 8.38 mg/kg (ZH) zilpaterol hydrochloride (ZH) daily for 21 d. Placebo or ZH was added to the diet 24 d, and short-term D was added 13 d before slaughter. Treatments were removed from all diets 3 d before slaughter. Plasma total calcium (Ca(2+)) was determined at study initiation, start of ZH and short-term D feedings, and at vitamin D3 and ZH withdrawal. Both plasma total and ionic Ca(2+) were determined when animals were sent to harvest. Longissimus muscle total and ionic Ca(2+) were determined in meat aged 7 and 4 d postmortem, respectively. When ZH was fed, long-term D decreased plasma total Ca(2+) at slaughter (P < 0.04). Short-term D increased (P < 0.01) plasma total and ionic Ca(2+) at slaughter regardless of ZH inclusion in the diet. Long- and short-term D, with or without ZH, did not affect (P > 0.28) LM total Ca(2+); however, both long- and short-term D increased LM ionic Ca(2+) when ZH was not fed (P < 0.01). Long-term D reduced LM ionic Ca(2+) when ZH was fed (P < 0.02). Neither long- nor short-term D affected PPARα or δ gene expression (P = 0.19) whether or not ZH was fed. Expression of MYH1 and 2A (P < 0.05) but not 2X (P = 0.21) was decreased in steers fed ZH. Long-term D had no effect on MYH2A expression (P = 0.21). Short-term D increased MYH2A expression when ZH was not fed (P < 0.03). Calpain mRNA tended to be lower in steers fed ZH (P = 0.09), but was not affected by long- or short-term D regardless of whether or not ZH was fed (P = 0.39). Expression of calpastatin did not differ with vitamin D supplementation (P = 0.35). In conclusion, ZH decreased oxidative myosin expression, and when combined with long-term D, ZH decreased LM ionic Ca(2+). Moreover, vitamin D3 supplementation did not increase calpain mRNA. These results help explain why vitamin D3 does not improve tenderness in steers fed ZH.

Milk is known to contain high concentrations of saturated fatty acids-such as palmitic (16:0), myristic (14:0), and lauric (12:0) acids-that can raise plasma cholesterol in humans, making their presence in milk undesirable. The main objective of our candidate gene study was to develop genetic markers that can be used to improve the healthfulness of bovine milk. The sterol regulatory element binding transcription factor 1 (SREBF1) known to regulate the transcription of lipogenic genes together with SREBF chaperone and insulin induced gene 1 were the candidate genes. The results showed significant association of the overall SREBF1 haplotypes with milk production and variations in lauric (12:0) and myristic (14:0) acid concentrations in milk. Haplotype H1 of SREBF1 was the most desirable to improve milk healthfulness because it was significantly associated with lower lauric (12:0) and myristic (14:0) acid concentrations compared with haplotype H3 of SREBF1, and lower lauric acid (12:0) concentration compared with haplotype H2 of SREBF1. Haplotype H1 of SREBF1, however, was significantly associated with lower milk production compared with haplotype H3 of SREBF1. We did not detect any significant associations between genetic polymorphisms in insulin induced gene 1 (INSIG1) and SREBF chaperone and milk fatty acid composition. In conclusion, genetic polymorphisms in SREBF1 can be used to develop genetic tools for the selection of animals producing milk with healthier fatty acid composition.

Dried distillers grains with solubles (DDGS) contain elevated concentrations of CP, oil, and S, which can negatively impact performance and carcass characteristics in steers weaned at 205 d of age. Early weaned (EW) cattle, however, consume less DM and require increased CP. Furthermore, the energy required to dispose of excess N may actually decrease excessive fat accumulation, which can occur in EW cattle. Thus, we hypothesized that feeding diets with increased concentrations of DDGS to EW steers for the first 99 d would decrease fat thickness and increase harvest weights, and would not inhibit performance or marbling deposition. To test this hypothesis, 90 Angus × Simmental steers (199.7 ± 12.2 kg) were weaned at 134 d of age (EW) and allotted to 3 high concentrate diets (20% corn silage) containing either 0%, 30%, or 60% DDGS (15.7%, 15.8% and 21.7% CP, respectively). Dietary treatments were fed for 99 d, after which steers were placed on a common diet containing no DDGS (12.9% CP) until harvest at a common weight of 599 kg. Concentration of dietary DDGS did not affect ADG, DMI, or G:F during the growing phase (P > 0.41), did not produce any carryover effects on ADG, DMI, or G:F during the finishing phase (P > 0.26), and resulted in similar overall performance (P > 0.52). Dressing percentage (P < 0.05), HCW (P = 0.06), fat thickness (P = 0.10), and % KPH (P = 0.08) responded quadratically to early DDGS supplementation, increasing from 0 to 30% DDGS inclusion and decreasing from 30 to 60% DDGS inclusion, respectively. Marbling score was not affected (P > 0.46) by DDGS inclusion, but there was a tendency (P = 0.08) for the ratio of subcutaneous (SC) to intramuscular (IM) fat to be altered by DDGS inclusion. The ratio of IM to SC fat decreased from 0 to 30% DDGS inclusion and increased from 30 to 60% DDGS inclusion. These data suggest that inclusion of increased concentrations of dietary DDGS early in the feedlot phase does not negatively impact growth and performance of EW cattle, but does partition energy from carcass to noncarcass components. However, increased concentrations of DDGS did not negatively impact IM fat deposition. Utilization of energy for disposal of excess N may be responsible for changes in carcass weight and SC fat deposition.

RESUMO

The objective of this study was to determine the influence of beef LM nutrient components on beef palatability traits and evaluate the impact of USDA quality grade on beef palatability. Longissimus muscle samples from related Angus cattle (n = 1,737) were obtained and fabricated into steaks for trained sensory panel, Warner-Bratzler shear force (WBSF), lipid oxidation measured by thiobarbituric acid reactive substances (TBARS), fatty acid, and mineral composition analysis. Pearson phenotypic correlations were obtained by the correlation procedure of SAS. Beef palatability data were analyzed by the GLM procedure of SAS with USDA quality grade as the main effect. Specific mineral concentrations did not demonstrate strong correlations with WBSF or sensory traits (r = -0.14 to 0.16). However, minerals appeared to have a stronger relationship with flavor; all minerals evaluated except Ca and Mn were positively correlated (P < 0.05) with beef flavor. Stearic acid (C18:0), C18:2, C20:4, and PUFA were negatively correlated (P < 0.05) with all 3 panelist tenderness traits (r = -0.09 to -0.22) and were positively correlated (P < 0.05) with WBSF (r = 0.09 to 0.15). The MUFA were positively correlated (P < 0.05) with panelist tenderness ratings (r = 0.07 to 0.10) and negatively associated (P < 0.05) with WBSF (r = -0.11). The strongest correlations with juiciness were negative relationships (P < 0.05) with C18:2, C18:3, C20:4, and PUFA (r = -0.08 to -0.20). Correlations with beef flavor were weak, but the strongest was a positive relationship with MUFA (r = 0.13). Quality grade affected (P < 0.05) WBSF, TBARS, and all trained sensory panel traits, except livery/metallic flavor. As quality grade increased, steaks were more tender (P < 0.05), as evidenced by both WBSF and sensory panel tenderness ratings. Prime steaks were rated juiciest (P < 0.05) by panelists, whereas Select and Low Choice were similarly rated below Top Choice for sustained juiciness. Quality grade influenced (P < 0.05) beef flavor, but not in a linear fashion. Although there were significant correlations, these results indicate tenderness, juiciness, and flavor are not strongly influenced by individual nutrient components in beef LM. Furthermore, the positive linear relationships between USDA quality grade and beef palatability traits suggest quality grade is still one of the most valuable tools available to predict beef tenderness.
